The proteolysis of immunoglobulin G with long-acting thyroid-stimulating activity.

چکیده
1. Proteolysis of immunoglobulin G with long-acting thyroid-stimulating activity with papain and pepsin shortened the duration of thyroid stimulation in mice prepared for the McKenzie (1958) bioassay. The time-course appeared to be related to molecular size. 2. The activity after proteolysis was recovered solely in the electrophoretically slow F(ab)-fragment. 3. There was no immunological cross-reaction between pituitary thyroid-stimulating hormone and the active fragments of the long-acting thyroid stimulator despite their similar behaviour in the bioassay. 4. The implications of these findings are discussed.

many as a single entity. The aetiology of the disease remains obscure. It has, however, been clearly established that thyroid-stimulating hormone (TSH) is not responsible for the excessive thyroid activity. An abnormal thyroid stimulator (known as long-acting thyroid stimulator, or LATS), which is both chemically and immunologically distinct from TSH, is found in the serum of many patients with...
متن کاملNeonatal hyperthyroidism and long-acting thyroid stimulator protector.
Neonatal hyperthyroidism has been thought to result from transplacental passage of long-acting thyroid stimulator (L.A.T.S.) from a mother with Graves's disease. A case is presented here in which no L.A.T.S. was detected in the mother or neonate but another immunoglobulin, L.A.T.S. protector, a specific human thyroid stimulator, was shown to be present in the mother's serum. This stimulator may...
متن کاملImmunoglobulin G inhibitor of thyroid-stimulating antibody is a cause of delay in the onset of neonatal Graves' disease.
Studies were carried out with the serum IgG from a mother and her two children who developed neonatal Graves' disease several weeks after birth. The maternal IgG: (a) stimulated the human thyroid in vitro, but maximal stimulation was found only with dilution of the IgG; (b) was very potent in the long-acting thyroid stimulator (LATS)-protector assay, but only when an inhibitor of the system was...
متن کاملThe effects of long term handling stress on thyroid function in male rats
Inappropriate responsiveness of the stress system may cause numerous alterations in endocrine, metabolic and autonomic systems. Many studies have been carried out on the effects of acute stress on thyroid function but the effects of chronic stress have not been fully investigated. The purpose of this study is to evaluate the effects of long term handling stress on thyroid function. Study was pe...
متن کاملInhibition of long-acting thyroid stimulator by thyroid particulate fractions.
The long-acting thyroid stimulator (LATS), found uniquely in the serum of patients with Graves' disease, is closely associated with the immunoglobulin G (IgG) proteins of serum and is removed from serum by antibody to human IgG (2, 3). Since some IgG molecules function as antibodies, it seems possible that LATS is also an antibody. This possibility would be strengthened if LATS could be demonst...
